CLEANING THE DOT-BLOTTER

Place dotter into glass dish and add ca. 500 ml of 0.5% SDS

Autoclave using liquid cycle for 15 minutes

After the dotter as cooled down pour off the 0.5 % SDS

Wash the dotter three times with DEPC treated water. Make sure to adequately flush the wells by moving the dotter up and down in the water 10 to 15 times

Wash the dotter with a small amount of ethanol

Dry under heat lamp

Pre-wet the charged nylon membrane and assemble dotter. The membrane needs to be pre-wetted (I use DEPC treated DI) or the dotter will not form a good seal.
